In meinem ersten Bericht zeigte ich bereits, dass der ER-Stresssensor IRE-1 im ? Germinal Center B Cell ( GCB ) Lymphoma ? weniger exprimiert wird als im ? Activated B Cell-Like ( ABC ) Lymphoma ?.

GCB und ABC sind zwei spezifische Untergruppen der DLBCL.

Folglich ist die Produktion des ?downstream?-liegenden Transkriptionsfaktors XBP1 in GCB Zelllinien nach Stimulation mit ER-Stress-induzierenden Faktoren erheblich niedriger.

Eine neuerliche Hypothese macht die Induktion der ER-Stresssignalwege durch HIV-PI dafür verantwortlich ( Abbildung ).

Resultate des ersten Jahres Um die ER-Stressantwort in DLBCL zu charakterisieren, wurden diese biochemisch in Tumorzelllinien der zwei bedeutenden Gruppen von DLBCL, ABC und Germinal Center B-Zell ( GCB ) DLBCL, analysiert.

Die Zelllinien wurden mit Tunicamycin oder Thapsigargin, zwei Molekülen, die ER-Stress auslösen, behandelt.

Diese ist durch eine Hochregulierung von UPR-abhängigen Genen, wie zum Beispiel DNAJB9, charakterisiert.

Diese vorläufigen Daten weisen darauf hin, dass der Verlust von IRE1 ein Kennzeichen von GCB DLBCL Tumoren ist und zur erhöhten Sensibilität gegenüber ER-Stress-induzierbaren Mitteln beitragen könnte.Möglicherweise lassen sich in Zukunft damit neue prognostische Werkzeuge und Therapien für DLBCL Patienten entwickeln.

Ausblick Um die physiologische Relevanz dieser Ergebnisse zu überprüfen, verfolgen wir vor allem zwei Strategien:

In the first year report, I showed that the ER-stress sensor IRE1 is downregulated in germinal center B cell ( GCB ) lymphoma when compared to activated B cell-like ( ABC ) lymphoma ;

these are two specific subsets of DLBCL.

Consequently the production of XBP1, a potent downstream transcription factor, was impaired in GCB cell lines after treatment with ER stress-inducing drugs.

Recently it has been suggested that HIV protease inhibitors, a family of drugs used to decrease viral replication in HIV patients, exert antitumor action by modulating a specific UPR response ( Figure ).

Results after the 1st year To characterize the UPR response in DLBCL, I biochemically analyzed the response in tumor cell lines originated from the two main subsets of DLBCL: activated B-cell-like ( ABC ) and germinal center B-cell-like ( GCB ) DLBCL.

I treated these cell lines with drugs that trigger ER stress such as tunicamycin and thapsigargin, or with the HIV protease inhibitor nelfinavir, and then monitored the activation of the UPR pathways.

Our results indicate that XBP1 reconstitution provides GCB cells with the capacity to mount a normal ER stress response characterized by an upregulation of the UPR-dependent genes, such as DNAJB9, upon treatment with ER stress inducers.

These preliminary data indicate that IRE1 deficiency is a hallmark of GCB DLBCL tumors and could contribute to increased sensitivity to ER stress-inducing drugs, possibly providing us with new prognostic tools and therapeutic strategies in DLBCL patients.
